The directory there is probably used only for the 'New mail'
notificationwhen the user logs in.
Clients such as mutt or pine (when they're accessing the local mail
store directly) you'll need to reconfigure via their configuration
files.
For POP-3 and IMAP clients, review the documentation of the server
software you're using. For POP-3, I'm using tpop3d which can be very
nicely configured via tpop3d.conf (and supports virtual users very
easily, so I recommend it very much). If you're using UW-IMAPD, you'll
probably have to patch the source -- they still haven't reached the age
of configuration files.
